Brown's studies on the mechanism of action of nuclear hormone receptors, including the estrogen (ER) and androgen (AR) receptor, has led to insights into the origin and treatment of human cancer. This work addresses the question as to how steroid receptors act as hormone-dependent transcription factors. He has dissected the molecular properties of ER associated co-regulatory proteins and their target gene network and the remodeling of this network by epigenetic changes during cancer development providing an understanding of the tissue selective mechanism of action of ER modulators widely used in the treatment of breast cancer. In 2016, Brown was elected to the National Academy of Sciences.
