With his lab colleagues, showed that specific components of bone are critical for the ordered regulation of blood stem cells and their ability to regenerate blood cell production after transplantation. Defined bone cells as key components of hematopoietic stem cell (HSC) niche. Discovered requirement for calcium sensing receptor and G proteins in HSC homing and bone marrow engraftment. Discovered parathyroid hormone's ability to modulate HSC niche and improve transplantation. Demonstrated role of p16ink4a in stem cell aging. Established the stem cell niche as a drug target and showed it can be manipulated by medicines to improve tissue regeneration, providing a paradigm for the development of regenerative therapies. Recognized the transformative potential of stem cell biology in medicine and created the multidisciplinary Center for Regenerative Medicine at Massachusetts General Hospital. With Douglas Melton, created the Harvard Stem Cell Institute.~~
