His laboratory was instrumental in the development and application of novel methods (e.g., optical tweezers, single-molecule fluorescence microscopy, scanning atomic force microscopy) for visualization, manipulation and experimental interrogation of biological macromolecules, in particular DNA-binding molecular motors, mechanical unfolding of globular proteins and RNAs, and the structure of chromatin and other protein-nucleic acid complexes.
